Can a man and a woman (both married and in love with their partners) be the best of friends without anyone doubting their friendship?

If you're leaving it open for other people to doubt the friendship, then I have to answer No, because inevitably SOME people (friends, SOs, etc) will say that it can't be and may think that something is up.
That aside, if you ask me personally, I think that yes, they can be friends, close friends even, but "the best of friends"...mmm...that might be a tad too close for comfort.

When I mean best friend I meant that they can discuss anything without being judged by the other. True friendship is unconditional, so who is to say that a man/woman cannot be friends?

Actually I have a friend I used to date. DH was nervous about meeting him- but when he did- he realized he is a cool guy. In fact, we're seeing him tomorrow. Not to mention I have a bunch of guy friends frmo college who are all married now- and we are still friends.

Discuss anything without being judge by the other - then yes.
However, I have to disagree with you that true friendship is unconditional. I think you can be a true friend but there may be a condition that would end that friendship.
I think the love between a parent and child is unconditional. Love of a family member, and even in some (certainly not all) marriages is unconditional.

Thank you all for your opinions. My male friend lives in another state and we communicate through email and phone. His wife and children are so very sweet and my husband has no worries about the friendship. I was just curious what you guys thought.

I have the same type of friendship with an old college friend. We see each other maybe once a year but we email each other almost everyday. He's about to get engaged. I think it's totally fine and DH is fine with it also.
I thought you meant a much closer friendship than that. I could see where others looking in might be judgemental about it or where, if one of the SOs had a problem with it, it could become a problem.

I think they can be friends, sure...but a best friend to me is someone that you have great confidence with, talk to constantly, call for opinions, hang out with often........that would be a problem.
