We were just there for a wedding in December and stayed at the Southernmost Point Hotel...it was at one end of Duval street and really nice and convenient to everything.
It's in Key West, BTW.

We've stayed at the sister one next door - Casa Marina and they are both really nice - the quiet side of Duvall, but not a bad stroll to the action.
If you want total seclusion Hawk's Cay is really nice.
I would be wary of other places on the other keys. Especially Marathon Key - they tend to be right on the road.

Islamorada and Marathon are both really nice and not as busy as Key West- but they're easily drivable to there....

We also stayed at the Wyndham Casa Marina and it is very nice. It's a long walk to Duval St so we drove or took a cab. But it's a nice hotel and in the middle of the day, the wait staff brings you ice pops by the pool!

I've been to Key West about a dozen times and prefer the guesthouses in Old Town to the large hotels. I love the personal touch at the guesthouses, plus they usually have a great breakfast and/or Happy Hour every day.
