It depends. FMU is the most concentrated since you haven't been drinking over night to dilute it. Sometimes, especially with the less sensitive tests or if you are very early in pregnancy, it helps the use FMU.
When I got my + with my daughter, I took my first test at night & the line was MUCH lighter than on the two tests I took early the next day.

Ya know, someone had mentioned a couple of weeks ago that your urine can be dilluted throughout the day, especially if you drink a lot of water. So I do believe that the best time of day is with first morning urine!
