Hotel -- Belhurst Castle.
Don't stay in the castle part, though. Stay in the Vinifera Inn part. It's the same building, but the Vinifera rooms are newer. They have hot tubs in the rooms (a little cheesy, I know) and fire places. Belhurst is also a winery. Their wine is okay, but overpriced for what it is. I think you get a free tasting with your stay. They have two restaurants at the hotel that are very nice -- one is a pub style and one is a little bit nicer. The food is great.
I'd recommend taking a wine tour. We used a fantastic company and it wasn't terribly expensive. I can look that up if you'd like the info. We've used them 3 times now and always had good luck with them.
Prejean, Fox Run, Wagner, and Heron Hill all have great wines. Definitely worth a visit! Hazlitt doesn't have the best wines, but it's probably the most fun winery to taste at. Definitely stop in there.
Their annual Deck the Halls celebration is coming up in November and December. That's always fun (you can google that to get more info!)
We go to the Fingerlakes as often as possible -- we love it up there. Enjoy!

The nicest area is around Seneca Lake, although there are portions around Candaigua Lake that are beautiful. I used to live near the latter before moving to LI.
Seneca Lake - there is Walkin's Glen on the south end and then Geneva up on the north end. There are winieries on both sides (east & west). My family goes wine tasting yearly and they love Glenora Wines - they have these huge picnic areas around the winery as well as huge (fake) wooden barrels to climb on and get fantastic views of the lake.
You really can't go wrong with any of the places to stay. If you want to camp, stay in Walkin's Glen State Park. There are also bed & breakfasts. As you go further into Geneva, it may get a little $$$$. It's considered a resort area for those people from the Rochester area who want a second home/summer home on the water. There are also the typical Holiday Inn, Hampton Inn, etc to stay at.
In Candaigua there is the Performing Arts center and a beautiful garden center to tour around, plus boat cruises, and shopping.

I stayed at the Mirbeau Inn and Spa near the Fingerlake Skaneateles...

That's just what I remember off the top of my head. I have a whole notebook at home of every vineyard we've been to up there... I, uh, have dozens more reviews.
ETA -- We're planning a little trip up there again soon and I'm going to look into the spa you posted!!

That's awesome! Thank you so much for posting! We now live upstate not too far from the Fingerlakes and a lot of our family is very into wine... It's hard because DH doesn't drink it so we are clueless on where to bring our guests!
The spa was fantastic! I would go back in a heartbeat!

DH hates wine to, but he doesn't mind going and seeing them.
