Not sure about a zoo in Lancaster, but if your son loves trains there is the Strasburg Railroad where you ride on an old steam engine, it's lots of fun. Then across the street is a Railroad Museum where there are a bunch of old trains, some you can even go inside, plus there is a little place where kids can play with toy trains.

Enjoy DW - we had so much fun when we went in early June. Didn't see any zoos to visit but we stayed on a farm (B&B) so didn't need a zoo - had plenty of animals there.
We did the children's museum - it is really adorable - we spend like an hour and a half there and really could have stayed longer - DS was having a blast. Also did the rr museum - not sure if your 16 mo old would like that. My 2 year old looooved it.
