If so what do you do? What do you say? Letter, email, telephone?

I always follow-up. If I have their number, I would call telling them I am just confirming that they received it... or if it was via email, I would send a short email, asking the same.

It can't hurt to follow up.
If they did receive your resume and just aren't interested, your follow-up won't put you any worse-off.
If by some fluke they didn't receive your resume and you're qualified for the job, it's a benefit to both you and the potential employer to make sure they have it. Maybe a phone call, if you have a contact name.
