Technically, I think the answer is immediately. Just expect to stop to feed/change them every 2-3 hours.

I found car rides to be easier when they were tiny -- I'd go upstate for 5-6hr drives when they were less than 8wks old and I knew they could sleep the whole time as long as I fed them every 2-3 hrs.

I'd be worried about exposing them to other people/germs in a rest stop until 4-6 weeks, though.

I'd say immediately.

I always hated how I could never see what was going on in the back of the car, so if I did a long trip like that, maybe have someone ride in the back next to baby.

Should be easy peasy, though. Newborns sleep like logs in the car!

I also agree with immediately. Do it when they are this small and sleep so much. It's sooooooooo much easier to do when they are in the newborn - 3 months range. The movement of the car will keep them sleeping for good stretches. Just have to stop every couple of hours for feedings & changing of diaper.

immediately.
However, I wouldn't do it. I'd wait at least 3 months...but that's just me. I have friends who have taken their 6 week old on an 11 hour plane ride!
Its up to you...but technically I don't think there is any restriction if the baby is healthy.
